The surname Hanke can be derived from three possible root words. In each instance, the name would have originated a patronymic name inspired by the personal name of the father. Firstly, Hanke can come from "Hanek," a Slavic short form for the name Johannes, from the Hebrew meaning "gracious gift of God." Or the name may come from a diminutive of the personal name Heinrich, from the Middle High German for "home-rule." Or it may derive from a number of ancient Germanic names beginning with the Old High German element "Hagan" meaning "enclosure." In this last case, the surname Hanke may also have been applied as a locative name, inspired by a geographical feature near where the bearer resided.

Early mentions of the name include Bartlome Hanngg, a resident of Ravensburg in 1480, and the Hanke family from Rehme who were granted a blazon of arms. Jacob Hennecke also known as Hencken or Hanke was a mayor in 1525. Hans Hanckin lived in Hochdorf in 1685, and Cord Hancke and his son Johann Hinrich Hanke in Rehme in 1665 and 1675.
